实验方案

  1. Sample Prep: Isolate PBMCs and resuspend in separation buffer.
  2. Labeling: Add the isolation cocktail to the sample and incubate for 5 minutes at room temperature.
  3. Bead Incubation: Add Anti-biotin Nanobeads and incubate for 10 minutes at 2-8°C.
  4. Separation: Apply the cell suspension to an LS Column placed in a magnetic field. Collect the flow-through, which contains the enriched, untouched CD4+ T cells.

What are the sample requirements?
This kit is optimized for use with Peripheral Blood Mononuclear Cells (PBMC). Fresh PBMCs yield the highest viability, but the kit is also compatible with cryopreserved PBMCs.
Can I stain the cells for Flow Cytometry immediately after isolation?
Yes. Because this is an "Untouched" (negative) selection kit, the target CD4+ T cells are free of bound antibodies or magnetic beads, leaving all surface markers available for staining.
How does the column-based negative selection work?
Non-CD4+ cells (CD8+ T cells, B cells, NK cells, Monocytes, etc.) are magnetically labeled. When passed through the column in a magnetic field, these labeled cells are retained, while the unlabeled CD4+ T cells pass through freely.
