Achieve superior enrichment of untouched TCR γδT cells, NK cells, or B cells by efficiently removing Human TCR alpha-beta (abT)+ Cells from PBMC. This Depletion kit utilizes a high-performance Column-based strategy combined with indirect Magnetic Beads labeling. By effectively trapping the unwanted αβ T cells in the column, the flow-through fraction contains the target cells of interest. 实验方案

  1. Preparation: Prepare a single-cell suspension from Human PBMCs.
  2. Labeling: Incubate cells with the primary antibody cocktail targeting TCR αβ.
  3. Bead Binding: Add the Anti-Biotin/Secondary Nanobeads to bind the primary antibodies.
  4. Separation: Apply the suspension to a column placed in a magnetic field.
  5. Collection (Depletion): The unwanted TCR αβ+ cells are retained in the column. Collect the flow-through, which contains the untouched target cells (e.g., γδT cells, NK cells).

What are the sample requirements?
This kit is optimized for fresh or cryopreserved Human PBMCs. It can also be used with Leukapheresis products. For frozen samples, ensure proper DNase treatment to prevent clumping.
Can I stain the isolated cells for Flow Cytometry immediately?
Yes. Since this is a depletion strategy, the cells in the flow-through (your target cells) have no magnetic beads or antibodies attached. Their surface markers are free and available for staining.
How does the Column-based separation work for depletion?
The column contains a ferromagnetic matrix. When placed in a magnetic field, it creates a high-gradient trap. This securely holds the magnetically labeled unwanted cells (TCR $αβ+$), preventing them from leaking into the flow-through, which ensures high purity of the non-magnetic fraction.
Are the separation columns reusable?
