实验方案

  1. Preparation: Prepare a single-cell suspension from Human PBMCs (fresh or frozen).
  2. Labeling: Add CD4 Microbeads directly to the cell suspension. Incubate to bind CD4+ T Cells.
  3. Separation: Place a separation column in the magnetic field and apply the cell suspension.
  4. Washing: Magnetically labeled CD4+ cells are retained in the column; unlabeled cells (CD8 T cells, B cells, etc.) are washed away.
  5. Elution: Remove the column from the magnet and elute the purified CD4+ T Cells.

Pour évaluer la pureté des cellules T CD4+ isolées, une séparation magnétique a été réalisée sur des cellules mononucléées du sang périphérique humain (PBMCs). Les cellules avant et après isolation ont été marquées avec des anticorps CD3-APC et CD4-PE et soumises à une analyse par cytométrie en flux. What are the sample requirements?
The kit is optimized for fresh or cryopreserved Human PBMCs. It is also highly effective with Leukapheresis products (Leukopaks). Ensure cells are well-dispersed and viable (>90%) for best results.
Do the magnetic beads affect cell activation?
No. The SOLIDEX® nanobeads are biodegradable and nano-sized. They are engineered to bind without inducing significant signal transduction or activation, ensuring the CD4+ cells remain phenotypically unaltered for functional assays.
How does the Column-based separation work?
The column contains a matrix of ferromagnetic spheres. When placed in a magnetic field, these spheres amplify the magnetic gradient, trapping the bead-labeled CD4+ cells. This strong retention allows for vigorous washing of contaminants before the target cells are released.
Are the separation columns reusable?
No. To maintain sterility and absolutely prevent cross-contamination between donor samples, the columns are designed for single-use only.
What is the advantage of Column-based over Column-free methods?
Column-based positive selection generally delivers higher purity. The physical column matrix allows for the washing away of non-specific cells that might get trapped in the "clumps" formed by column-free tube methods.
