实验方案

  1. Preparation: Prepare a single-cell suspension from cryopreserved or fresh PBMCs.
  2. Labeling: Add the Biotin-Antibody Cocktail (targets non-CD14 cells: T cells, B cells, NK cells, etc.). Incubate.
  3. Bead Binding: Add Anti-Biotin Microbeads to bind the labeled non-target cells. Incubate.
  4. Separation: Apply the suspension to the separation column placed in a magnetic field.
  5. Elution: Unlabeled CD14+ Classical Monocytes pass through the column freely and are collected (Untouched).

常见问题

What are the sample requirements?
We recommend using fresh or cryopreserved Human PBMCs. For cryopreserved samples, ensure high viability (>90%) and proper DNase treatment during thawing to prevent clumping. Leukapheresis products are also compatible.
Can I stain the cells for Flow Cytometry immediately after isolation?
Yes. Since this is an "Untouched" (Negative Selection) kit, the CD14 receptors and other surface markers are free and not blocked by magnetic beads or antibodies. You can proceed directly to staining.
How does the Column-based separation work?
The column contains a ferromagnetic matrix (spheres). When placed in a magnetic field, the matrix amplifies the magnetic force by 10,000-fold, effectively retaining the magnetically labeled non-target cells while unlabeled monocytes pass through.
Are the separation columns reusable?
No. The columns are designed for single-use to ensure sterility and prevent cross-contamination between patient samples.
What is the advantage of Column-based over Column-free methods?
Column-based methods typically offer higher purity. The column allows for washing of the retained cell mass, ensuring that only the specific "Untouched" cells elute.
