实验方案

  1. Preparation: Prepare a single-cell suspension from Human PBMCs (fresh or frozen).
  2. Labeling: Add CD8 Microbeads directly to the cell suspension. Incubate to bind CD8+ T Cells.
  3. Separation: Place a separation column in the magnetic field and apply the cell suspension.
  4. Washing: The magnetically labeled CD8+ cells are retained in the column; unlabeled cells (CD4+ T cells, B cells, Monocytes) are washed away.
  5. Elution: Remove the column from the magnet and elute the highly purified CD8+ T Cells.

Pour déterminer la pureté des cellules T CD8+ isolées, les cellules T CD8+ ont été triées magnétiquement à partir de cellules mononucléées du sang périphérique humain (PBMC). Les cellules avant et après séparation ont été marquées avec des anticorps CD3-APC et CD8-PE puis analysées par cytométrie en flux. Les résultats ont montré une augmentation remarquable de la pureté des cellules T CD8+ de 16,6 % avant isolation à 94,6 % après isolation, confirmant un enrichissement très efficace des cellules T CD8+.

What are the sample requirements?
This kit is optimized for fresh or cryopreserved Human PBMCs. It is also fully compatible with Leukapheresis products (Leukopaks). Ensure high cell viability (>90%) and proper DNase treatment if using frozen samples to prevent clumping.
Do the magnetic beads affect cell activation?
No. The SOLIDEX® nanobeads are biodegradable and extremely small (nano-scale). They are designed to minimize receptor cross-linking and avoid inducing artificial activation, ensuring your CD8+ cells remain in a physiological state for downstream functional assays.
How does the Column-based separation work?
